RESUMO

Root extracts from Danube Delta Nymphaea alba were used to prepare gold nanoparticles (AuNPRn) by reducing HAuCl4 at different pHs (6.4-8.4) using ultrasonic irradiation: an easy, cheap, eco-friendly and green approach. Their antibacterial and anticancer activities were evaluated against Staphylococcus aureus and Escherichia coli, and A2780 ovarian cancer cells, respectively. The AuNPRn were characterized concerning their phytoconstituents (polyphenols, flavonoids and condensed tannins) and gold content. All of the nanoparticles were negatively charged. AuNPRn exhibited a hydrodynamic size distribution ranging from 32 nm to 280 nm, with the larger nanoparticles being obtained with an Au/root extract ratio of 0.56, pH 7 and 10 min of sonication (AuNPR1), whereas the smallest were obtained with an Au/root extract ratio of 0.24, pH 7.8 and 40 min of sonication (AuNPR4). The TEM/SEM images showed that the AuNPRn had different shapes. The ATR-FTIR indicated that AuNPRn interact mainly with hydroxyl groups present in the polyphenol compounds, which also confirm their high antioxidant capacity, except for AuNPR2 obtained at pH 6.4. Among the AuNPRn, the smallest ones exhibited enhanced antimicrobial and anticancer activities.

Seeking to enrich the yet less explored field of scorpionate complexes bearing antioxidant properties, we, here, report on the synthesis, characterization and assessment of the antioxidant activity of new complexes derived from three scorpionate ligands. The interaction between the scorpionate ligands thallium(I) hydrotris(5-methyl-indazolyl)borate (TlTp4Bo,5Me), thallium(I) hydrotris(4,5-dihydro-2H-benzo[g]indazolyl)borate (TlTpa) and potassium hydrotris(3-tert-butyl- pyrazolyl)borate (KTptBu), and metal(II) chlorides, in dichloromethane at room temperature, produced a new family of complexes having the stoichiometric formula [M(Tp4Bo,5Me)2] (M = Cu, 1; Zn, 4; Cd, 7), [M(Tpa)2] (M = Cu, 2; Zn, 5; Cd, 8), [Cu(HpztBu)3Cl2] (3), [Zn(TptBu)Cl] (6) and [Cd(BptBu)(HpztBu)Cl] (9). The obtained metal complexes were characterized by Fourier transform infrared spectroscopy, proton nuclear magnetic resonance and elemental analysis, highlighting the total and partial hydrolysis of the scorpionate ligand TptBu during the synthesis of the Cu(II) complex 3 and the Cd(II) complex 9, respectively. An assessment of the antioxidant activity of the obtained metal complexes was performed through both enzymatic and non-enzymatic assays against 1,1-diphenyl-2-picryl- hydrazyl (DPPH·), 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+·), hydroxyl (HO·), nitric oxide (NO·), superoxide (O2-) and peroxide (OOH·) radicals. In particular, the complex [Cu(Tpa)2]⋅0.5H2O (2) exhibited significant antioxidant activity, as good and specific activity against superoxide (O2-·), (IC50 values equal to 5.6 ± 0.2 µM) and might be identified as auspicious SOD-mimics (SOD = superoxide dismutase).

Surface modification of zinc oxide nanoparticles (ZnO NPs) is a strategy to tune their biocompatibility. Herein we report on the synthesis of a series of fluorescent ZnO NPs modified with 2-10% (3-glycidyloxypropyl)trimethoxysilane (GPTMS) to investigate the fluorescence properties and to explore their applications in microbiology and biomedicine. The obtained ZnO NPs were characterized by X-ray diffraction (XRD), high resolution transmission electron microscopy (HRTEM) and Fourier transform infrared spectroscopy (FTIR). Size reduction occurred from ca. 13 nm in unmodified ZnO to 3-4 nm in silane-modified samples and fluorescence spectra showed size-dependent variation of the photoemission bands' intensity. The antibacterial and cytotoxic activities were investigated on Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria, and in ovarian (A2780) and prostate (PC3) cancer cells by tetrazolium/formazan-based methods. The antibacterial effect was higher for E. coli than S. aureus, while the cytotoxic activity was similar for both cancer cells and varied with the particle size. Cell death by apoptosis, and/or necrosis versus autophagy, were explored by flow cytometry using an Annexin V based-method and transmission electron microscopy (TEM). The main mechanism of ZnO NPs toxicity may involve the generation of reactive oxygen species (ROS) and the induction of apoptosis or autophagy. This work revealed the potential utility of GPTMS-modified ZnO NPs in the treatment of bacterial infection and cancer.

In the last two decades, a tremendous amount of attention has been directed towards the design of antibacterial silver(I)-based materials, including coordination polymers (CPs) built up with a great variety of oxygen and nitrogen-containing ligands. Herein, a family of six new silver(I)-based CPs, having the general stoechiometric formula [Ag(H2DMPMB)(X)] (X = NO3, 1; CF3CO2, 2; CF3SO3, 3; BF4, 4; ClO4, 5; and PF6, 6) and incorporating the flexible ditopic pyrazolyl-type ligand 4,4'-bis((3,5-dimethyl-1H-pyrazol-4-yl)methyl)biphenyl (H2DMPMB), has been prepared by the chemical precipitation method involving the reaction of silver(I) salts with H2DMPMB in the 1:1 molar ratio, in alcohols, or acetonitrile at room temperature for two-hours. The new silver(I)-based polymeric materials were characterized by means of Fourier transform infrared spectroscopy (FTIR), elemental analysis (EA), and thermogravimetric analysis (TGA), allowing for the proposition that their structures comprise one-dimensional chains, with the silver(I) ions mostly assuming a T-shapped stereochemistry completed by the exo-bidentate ligands and counter-anions. The obtained silver(I) CPs showed a remarkable light insensitivity and stability in the air, are insoluble in water and in most common organic solvents, and possess appreciable thermal stabilities spanning the range 250-350 °C. The antibacterial activity of the obtained silver(I) CPs was tested against the Gram-negative bacteria Escherichia coli (E. coli) and Gram-positive bacteria Staphylococcus aureus (S. aureus) using the Tetrazolium/Formazan test (TTC), by measuring the bacterial viability at different time intervals. The complete reduction of both bacterial strains occurred after 24 h of exposure to all silver(I) CPs, the bacterial viability values for S. aureus reaching 8% for compounds 3, 5, and 6 after only two-hours.

A family of fifteen quaternary ammonium salts (QAs), bearing the 1,2-bis(4-pyridyl)ethane core, were obtained using for the first time two different green methods, such as microwave (MW) and ultrasounds (US) irradiation, with very good yields and in much shorter times compared to the classical method, and an assay on their antimicrobial action against Escherichia coli (E. coli) was carried out. While 12 to 24 hours were required for complete alkylation of 1,2-bis(4-pyridyl)ethane by reactive halogenated derivatives in anhydrous solvent under reflux conditions, MW and US irradiation reduced the reaction time and the desired products were achieved in a few min. One of the aims of this study was to evaluate the antibacterial potential of the synthesized QAs against pathogenic bacteria, along with their impact on germination activity of wheat seeds (Triticum aestivum L.). The antibacterial activity of the QAs against Escherichia coli was explored by determining the minimum inhibitory concentration (MIC). The MIC values varied from 0.312 to 2.5 mg/mL, highlighting the lowest values attained for the derivatives containing methoxy, chlorine and benzofurane functional groups. The viability of aerobic bacteria was determined with the Tetrazolium/Formazan Test, a method that was found to be the best alternative approach with respect to the difuzimetric method. Seeds of Triticum aestivum L. were used for the evaluation of the germination indicators, such as seed germination (SG), the relative seed germination (RSG), the relative radicle growth (RRG), and the seed germination index (GI). The toxicity studies of QAs 1, 4 and 7, at two different concentrations, showed no inhibitory effect on seed germination.

The comparative effect of two quaternary ammonium salts from 1,2-bis(4-pyridyl)ethane (PyQAs), namely N,N'-diphenacyl-1,2-bis(4-pyridinium)ethane dibromide (PyQAs1) and N,N'-di(p-methoxyphenacyl)-1,2-bis(4-pyridinium)ethane dibromide (PyQAs2), upon the size and photoluminescence of zinc oxide nanoparticles (ZnO NPs) was investigated. The formation of ZnO NPs took place in the presence of variable amounts of the two PyQAs species (1, 2.5, and 5%), according to the chemical precipitation of zinc(II) acetate with potassium hydroxide in ethanol under reflux. The obtained ZnO NPs were structurally characterized by means of X-ray powder diffraction, infrared, and Raman spectroscopy. The fluorescence of all supernatant solutions, observed under ultraviolet light, determined us to make an investigation of the solutions by means of liquid chromatography coupled with electrospray ionization mass spectrometry (LC-MS-ESI) in order to elucidate the identity of the newly formed fluorescent species. Such an occurrence thus allowed the invocation of the catalytic effect of zinc(II) ions towards the organic transformation of both nonfluorescent PyQAs surfactants into new fluorescent organic species.

The five metal azolate/carboxylate (MAC) compounds [Cd(dmpzc)(DMF)(H2O)] (Cd-dmpzc), [Pd(H2dmpzc)2Cl2] (Pd-dmpzc), [Cu(Hdmpzc)2] (Cu-dmpzc), [Zn4O(dmpzc)3]·Solv (Zn-dmpzc·S), and [Co4O(dmpzc)3]·Solv (Co-dmpzc·S) were isolated by coupling 3,5-dimethyl-1H-pyrazol-4-carboxylic acid (H2dmpzc) to cadmium(II), palladium(II), copper(II), zinc(II), and cobalt(II) salts. While Cd-dmpzc and Pd-dmpzc had never been prepared in the past, for Cu-dmpzc, Zn-dmpzc·S, and Co-dmpzc·S we optimized alternative synthetic paths that, in the case of the copper(II) and cobalt(II) derivatives, are faster and grant higher yields than the previously reported ones. The crystal structure details were determined ab initio (Cd-dmpzc and Pd-dmpzc) or refined (Cu-dmpzc, Zn-dmpzc·S, and Co-dmpzc·S) by means of powder X-ray diffraction (PXRD). While Cd-dmpzc is a nonporous 3D MAC framework, Pd-dmpzc shows a 3D hybrid coordination/hydrogen-bonded network, in which Pd(H2dmpzc)2Cl2 monomers are present. The thermal behavior of the five MAC compounds was investigated by coupling thermal analysis to variable-temperature PXRD. Their catalytic activity was assessed in oxidative and C-C coupling reactions, with the copper(II) and cadmium(II) derivatives being the first nonporous MAC frameworks to be tested as catalysts. Cu-dmpzc is the most active catalyst in the partial oxidation of cyclohexane by tert-butyl hydroperoxide in acetonitrile (yields up to 12% after 9 h) and is remarkably active in the solvent-free microwave-assisted oxidation of 1-phenylethanol to acetophenone (yields up to 99% at 120 °C in only 0.5 h). On the other hand, activated Zn-dmpzc·S (Zn-dmpzc) is the most active catalyst in the Henry C-C coupling reaction of aromatic aldehydes with nitroethane, showing appreciable diastereoselectivity toward the syn-nitroalkanol isomer (syn:anti selectivity up to 79:21).

Adducts of a number of tertiary pnicogen ligands ER(3) (triphenyl-phosphine and -arsine (PPh(3),AsPh(3)), diphenyl,2-pyridylphosphine (PPh(2)py), tris(4-fluorophenyl)phosphine (P(C(6)H(4)-4F)(3)), tris(2-tolyl)phosphine (P(o-tol)(3)), tris(cyclohexyl)phosphine (PCy(3))), with silver(I) thiocyanate, AgSCN are structurally and spectroscopically characterized. The 1:3 AgSCN : ER(3) complexes structurally defined (for PPh(3),AsPh(3) (diversely solvated)) take the form [(R(3)E)(3)AgX], the thiocyanate X = NCS being N-bound, thus [(Ph(3)E)Ag(NCS)]. A 1:2 complex with PPh(2)py, takes the binuclear form [(pyPh(2)P)(2)Ag()Ag(PPh(2)py)(2)] with an eight-membered cyclic core. 1:1 complexes are defined with PPh(2)py, P(o-tol)(3) and PCy(3); binuclear forms [(R(3)P)Ag()Ag(PR(3))] are obtained with P(o-tol)(3) (two polymorphs), while novel isomeric tetranuclear forms, which may be envisaged as dimers of dimers, are obtained with PPh(2)py, and, as further polymorphs, with PCy(3); these latter may be considered as extensions of the 'cubane' and 'step' forms previously described for [(R(3)E)AgX](4) (X = halide) complexes. Solvent-assisted mechanochemical or solvent-assisted solid-state synthesis methods were employed in some cases, where complexes could not be obtained by conventional solution methods, or where such methods yielded a mixture of polymorphs unsuitable for solid-state spectroscopy. The wavenumbers of the ν(CN) bands in the IR spectra are in broad agreement with the empirical rule that distinguishes bridging from terminal bonding, but exceptions occur for compounds that have a double SCN bridged dimeric structure, and replacement of PPh(3) with PPh(2)py apparently causes a significant decrease in ν(CN) to well below the range expected for bridging SCN in these structures. (31)P CP MAS NMR spectra yield additional parameters that allow a correlation between the structures and spectra.

Coupling the rigid spacer 4,4'-bipyrazole (H(2)BPZ), in its anionic or neutral form, to different silver(I) salts allowed isolation of the novel coordination polymers [Ag(2)(BPZ)] (1) and [Ag(H(2)BPZ)(X)] (X = NO(3), 2; ClO(4), 3; BF(4), 4; PF(6), 5; CH(3)SO(3), 6; CF(3)SO(3), 7), which were fully characterized by infrared and emission spectroscopies, thermal analysis, and X-ray powder diffraction. The crystal structure of 1 consists of 2-D layers containing 1-D chains of Ag(I) ions bridged by exo-tetradentate bipyrazolato moieties. The crystal structures of the [Ag(H(2)BPZ)(X)] species 2-7 feature 1-D chains of [Ag(H(2)BPZ)] stoichiometry, along which the metal centers are bridged by exo-bidentate bipyrazolyl spacers. Contacts among adjacent chains are mediated by the counterions through nonbonding interactions involving the Ag(I) ions and the pyrazolyl N-H groups. Thermogravimetric analyses disclosed the good thermal stability of these materials, decomposing in the range 200-300 °C. Under UV irradiation at room temperature, all the species showed a yellow-green emission centered in the range 520-522 nm. When embedded into polyethylene disks, 1, 2, and 4-7 demonstrated their activity as topical antibacterial agents against suspensions of E. coli, P. aeruginosa, and S. aureus: complete reduction of the three bacterial strains was achieved in 24 h, reduction of S. aureus reaching ca. 90% in only 2 h. Biocidal action was expressed also by contact susceptibility tests.

A number of adducts of copper(I) thiocyanate with bulky tertiary phosphine ligands, and some nitrogen-base solvates, were synthesized and structurally and spectroscopically characterised. CuSCN:PCy3 (1:2), as crystallized from pyridine, is shown by a single crystal X-ray study to be a one-dimensional polymer ...(Cy3P)2CuSCN(Cy3P)2CuSCN... (1) with the four-coordinate copper atoms linked end-on by S-SCN-N bridging thiocyanate groups. A second form (2), obtained from acetonitrile, was also identified and shown by IR and 31P CPMAS NMR spectroscopy to be mononuclear, with the magnitude of the dν(Cu) parameter measured from the 31P CPMAS and the ν(CN) value from the IR clearly establishing this compound as three-coordinate [(Cy3P)2CuNCS]. Two further CuSCN/PCy3 compounds CuSCN:PCy3 (1:1) (3), and CuSCN:PCy3:py (1:1:1) (4) were also characterized spectroscopically, with the dν(Cu) parameters indicating three- and four-coordinate copper sites, respectively. Attempts to obtain a 1:2 adduct with tri-t-butylphosphine have yielded, from pyridine, the 1:1 adduct as a dimer [(Bu(t)3P)((SCN)(NCS))Cu(PBu(t)3)] (5), while similar attempts with tri-o-tolylphosphine (from acetonitrile and pyridine (= L)) resulted in solvated 1:1:1 CuSCN:P(o-tol)3:L forms as dimeric [{(o-tol)3P}LCu((SCN)(NCS))CuL{P(o-tol)3}] (6 and 8). The solvent-free 1:1 CuSCN:P(o-tol)3 adduct (7), obtained by desolvation of 6, was characterized spectroscopically and dν(Cu) measurements from the 31P CPMAS NMR data are consistent with the decrease in coordination number of the copper atom from four (for 6) (P,N(MeCN)Cu,S,N) to three (for 7) (PCuS,N) upon loss of the acetonitrile of solvation. These results are compared with those previously reported for mononuclear and binuclear PPh3 adducts which demonstrate a clear tendency for the copper centre to remain four-coordinate. The IR spectroscopic measurements on these compounds show that bands in the far-IR spectra provide a much more definitive criterion for distinguishing between bridging and terminal bonding than does an often-used empirical rule based on ν(CN) in the mid-IR, which leads to the wrong conclusion in some cases.

The reaction of the rigid spacer 4,4'-bipyrazole (H(2)BPZ) with late transition metals, either following conventional routes or under solvothermal conditions, afforded the coordination polymers [M(BPZ)]·Solv (M = Zn, 1; Co, 2; Cd, 3; Hg, 4; Cu, 5; Ni, 6; Pd, 7; Solv = DMF, 3; MeCN, 5 and 6; H(2)O, 7), [Cu(H(2)BPZ)(2)(NO(3))(2)] (8), and [Cd(H(2)BPZ)(CH(3)COO)(2)] (9). State-of-the-art laboratory powder diffraction methods allowed to disclose the isomorphous character of 1 and 2, as well as of 5 and 6, which feature 3D porous networks containing 1D channels of square and rhombic shape, respectively. 3, crystallizing in the relatively rare P6(1)22 space group, consists of homochiral helices of octahedral Cd(II) ions, packing in bundles mutually linked by "radial", nonplanar BPZ ligands. Finally, the dense species 8 and 9 contain parallel 2D layers of square and rectangular meshes, respectively. Thermogravimetric analyses witnessed the relevant thermal robustness of all the [M(BPZ)] materials [except the mercury(II) derivative], which are stable in air at least up to 300 °C, with the zinc(II) derivative decomposing only around 450 °C. Variable-temperature powder diffraction experiments highlighted the permanent porosity of 1-3, 5, and 6, retained along consecutive temperature cycles in all cases but 3. When probed with N(2) at 77 K, 1-3 and 5-7 showed Brunauer-Emmett-Teller and Langmuir specific surface areas in the ranges 314(2)-993(11) and 509(16)-1105(1) m(2)/g, respectively.

Solvothermal reactions of the flexible, pyrazole-based 1,4-bis((3,5-dimethyl-1H-pyrazol-4-yl)methyl)benzene ligand (H(2)BDMPX) with late transition metal ions allowed the isolation of the four coordination compounds M(x)(BDMPX) (x = 1 for M = Zn, 1; Co, 2; Cd, 3; x = 2 for M = Cu, 4). The investigation of the thermal behavior assessed the high thermal robustness of these materials, which are stable in air at least up to 300 °C, with the Cd(II) derivative starting to decompose only around 500 °C. As retrieved by ab initio X-ray powder diffraction, the isomorphous compounds 1-3 possess a dense 3-D network featuring rhombic motifs hinged about rigid and parallel chains of tetrahedral MN(4) chromophores. As demonstrated by thermodiffractometric measurements, temperature increase triggers framework flexibility. The latter is at work also when N(2) adsorption is assayed at 77 K: 1 and 2 show permanent porosity, with BET and Langmuir specific surface areas of 515, 667 m(2)/g and 209, 384 m(2)/g, respectively. 1 and 2 thus represent an intriguing example of "porosity without pores", their pro-porous nature being explained in terms of the flexibility of the rhombic motifs, stimulated by the gas probe and facilitated by the nature of the ligand.
